In lab, our attempt to clone BBa_J100249 was unsuccessful. There was no distinguishable difference between colonies grown in a negative control and colonies with the promoter. If the promoter was successfully implemented, (1) at least some of the colonies on the promoter tray would be clear because they did not produce green fluorescent protein. If this promoter is imperfect, we (2) might have expected to see some colonies that produced red fluorescent protein. Neither of these scenarios were observed in the promoter tray.
